Donor Urine Variation Influenced the Hydrolysis Protocol

The developers did not choose the final urine incubation time solely from a favorable average. Testing different donors’ urine revealed variation in THC-COOH-glucuronide cleavage that informed the final procedure.

The donor challenge added information

In the assay-development work, urine from five donors was fortified with THC-COOH-glucuronide and incubated with beta-glucuronidase for half an hour. Mean cleavage was approximately 90%, but the lowest observed cleavage was 74%. The paper identified urine dilution and pH variation as reasons to consider specimen differences.

Longer incubation was a procedural choice

The authors reported that longer incubation was not associated with loss of signal and selected a final one-hour incubation, adding a half-hour safety margin. That decision does not demonstrate that every possible future urine specimen will achieve exactly the same cleavage. It does show why a single preparation condition should be supported with more than one convenient specimen.

What to review in another laboratory

Relevant records include enzyme source, incubation duration and temperature, matrix challenges and the conjugated control actually used. This is intentional hydrolysis in urine, distinct from unwanted cleavage when measuring free metabolite in blood. A preparation-dependent metabolite result should not be interpreted as a direct measure of behavioral impairment or converted into an exact cannabis dose.

Repeated Oral THC Increased Daily Dose by Changing Administration Frequency

The intensive oral-THC regimen repeatedly used 20 mg capsules while shortening the intervals between administrations. A higher daily total did not mean every individual capsule dose became larger.
