Can Portable Brain Imaging Measure THC Intoxication? What Prefrontal Connectivity Shows

Oral THC altered prefrontal brain signals in a controlled human experiment, but those changes do not yet establish a universal roadside impairment test. The distinction is between a reproducible research association and a validated individual decision rule.

What did the instrument record?

Karunakaran and colleagues used functional near-infrared spectroscopy, an optical technique sensitive to blood-oxygenation changes, during resting-state assessments. Adults who regularly used cannabis received oral THC and placebo in randomized order. Of 185 randomized participants, 128 completed both days with usable data.

THC doses were individualized to produce intoxication despite tolerance. The study therefore did not test one identical dose across all participants or measure ordinary undisturbed roadside behavior.

Connectivity and subjective effects

THC increased subjective intoxication and heart rate. Prefrontal connections became less stable, and static correlations and anticorrelations changed relative to placebo. Some connectivity measures were associated with greater reported intoxication.

These optical signals are not direct measurements of THC concentration or recordings of every neuron. Their interpretation depends on acquisition, signal processing, the brain regions sampled, and physiological influences on the measurements.

What would an individual test require?

A group-level change does not supply sensitivity, specificity, and error rates for every proposed field application. A usable impairment classifier would need an explicit decision rule and validation in the intended population and setting, including people with other causes of altered physiology or performance.

The paper provides evidence that acute THC exposure can disrupt resting prefrontal dynamics. It does not establish that a particular optical pattern proves unsafe driving, permanent brain injury, or impairment caused exclusively by cannabis. An expert should identify the actual endpoint before translating a brain-imaging result into a behavioral conclusion.

Can THC Analgesia Be Separated From Memory Effects? A Peptide Experiment in Mice

An experimental peptide preserved THC-related analgesic responses while preventing a memory-task deficit in mice. The result is a preclinical drug-development finding, not evidence that an available supplement makes cannabis use free of cognitive effects. Targeting a receptor interaction Gallo and colleagues investigated peptides designed to disrupt an interaction between cannabinoid…

Can Cannabis Users Self-Titrate Across Different Potencies? What Blood Verification Adds

Different amounts of cannabis flower did not necessarily produce different measured THC-related blood exposure in a preliminary naturalistic study. The finding supports investigating self-titration, rather than assuming product potency alone determines absorbed dose. A three-day product assignment Bidwell and colleagues enrolled 24 male cannabis users and asked them to use…
