CB1 Receptor Density and Function Were Different Measurements in a Rat THC Study
A rat study found relatively few baseline sex differences in CB1 receptor density, yet clearer differences in receptor adaptation after repeated THC. Receptor number and receptor signaling should not be treated as interchangeable endpoints.
Brain tissue after a controlled regimen
Male and female rats underwent gonadectomy or comparison surgery, with selected hormone-replacement conditions. After repeated THC or vehicle, researchers examined four brain regions using receptor-binding and G-protein activation assays. These were tissue experiments, not human blood tests.
Baseline and post-treatment questions differed
Before repeated THC effects were considered, receptor-density differences between sexes were generally limited. One functional exception involved stronger hippocampal receptor activation in females. Repeated THC then produced downregulation and desensitization in both sexes, with greater changes in females on selected regional measures.
Regional exceptions mattered
The striatal findings did not show an identical sex difference for every measure: desensitization differed, while downregulation did not show the same sex-specific pattern. A broad statement that all female receptor measurements changed more would therefore be inaccurate.
No direct human hormone rule
Gonadectomy, hormone replacement, injected dosing and ex vivo assays do not establish how a patient’s hormonal status affects cannabis response. The study did not validate a clinical tolerance biomarker or a rule for adjusting THC doses. Its contribution is to distinguish receptor abundance, coupling, treatment history and brain region when interpreting proposed mechanisms.
