CB2 Regulation Did Not Follow a Simple Kinase-Recruitment Rule in Cells

Adding regulatory kinases did not substantially increase CB2 arrestin recruitment in an engineered-cell study. A familiar receptor-regulation model was not automatically applicable to this cannabinoid receptor.

Real-time cellular measurements

Researchers used CB2-expressing HEK293 cells and BRET assays to assess arrestin movement and G-protein signaling. They varied kinase expression and mutated selected receptor residues. These were in-vitro measurements, not animal behavior or patient tolerance.

Negative and positive findings differed

Overexpressing the tested GRK isoforms failed to produce a substantial recruitment enhancement, and inhibiting endogenous GRK2/3 did not substantially suppress arrestin translocation. Mutating C-terminal aspartic-acid residues, however, reduced arrestin recruitment and signaling desensitization.

The model suggests rather than settles

The results supported a possible phospho-mimic role for those residues. Engineered receptor abundance and the host-cell environment limit transfer to native tissues. A null kinase effect does not prove that phosphorylation never matters, nor does short-term desensitization directly measure clinical cannabis tolerance. The paper provides a testable regulatory mechanism while cautioning against importing CB1 or other receptor assumptions wholesale into CB2 biology.

Saliva Cannabis Detection Windows Change With THC AND Versus OR a Metabolite

Requiring both THC and its metabolite to exceed a cutoff can produce a different saliva detection window from accepting either compound. A controlled human cannabis-smoking study demonstrated that difference, particularly among occasional smokers. The result concerns how a test defines a positive sample; it does not establish how long a…

A CB2 Knockout Experiment Did Not Test Whether Cannabis Causes Psychosis

A mouse experiment linked altered cannabinoid CB2 receptor function with methamphetamine-related behavioral responses. It did not administer cannabis or determine whether THC exposure caused a psychiatric disorder. A Prenatal Alcohol Study Linked Exposure to Lifetime Cannabis Use, Not Earlier Initiation

A longitudinal study associated maternal alcohol dose per drinking occasion during pregnancy with greater reported lifetime cannabis use in young adulthood. It did not find the same association with every cannabis-use measure, including age at first use. Exposure preceded the later reports The analysis involved 121 young adults from the…
