Cannabis Metabolomics Associations Used Baseline and Placebo Differences Rather Than a Standalone Blood Value

The metabolomics study related cannabis effects to changes from baseline and differences from placebo. Those analyses did not directly validate classification from one isolated post-use serum sample.

The investigators constructed different change measures

For attentional lapses and subjective high, they subtracted the participant’s placebo-condition score from the cannabis-condition score. For metabolomics, they calculated changes from pre-dose baseline. Their models then examined associations between these defined changes, with additional participant characteristics included as predictors.

A change score needs a reference

An absolute concentration combines the person’s starting value with subsequent influences. A within-person change addresses a different question. If a real-world test lacks a pre-use sample or placebo comparison, it cannot simply reproduce the study’s calculations. The authors explicitly identify standalone sampling and full time-course validation as future research needs.

Association is a step toward validation

The controlled design helps investigate candidate relationships under known exposure conditions. It does not establish a ready-to-use decision rule or show that a later recovered sample is an interchangeable baseline. That substitution was discussed as a possibility requiring study. For interpretation, specify which reference values supported the association and which would be unavailable in the proposed application. This prevents a controlled change analysis from being presented as an already validated single-sample impairment assay.

The CBD Scan Trial Used Equal Allocation Despite Planning a Two-to-One Ratio

The scan-anxiety study ultimately randomized twenty-five participants to each treatment. The paper disclosed that its original sample-size plan anticipated a two-to-one allocation instead. The discrepancy concerned the allocation ratio The central registrar inadvertently implemented a one-to-one scheme rather than the originally intended unequal ratio. Participants still underwent randomized assignment, and…

Only Six Participants Had Severe Baseline Anxiety in the CBD Scan Trial

The scan-anxiety trial stratified randomization by baseline anxiety severity, but only six participants were in its severe category. Stratification did not supply enough data for a reliable severe-anxiety subgroup analysis. The strata helped organize allocation Participants were assigned within GAD-7 categories covering mild-to-moderate and severe anxiety. Of the fifty women…

The CBD Scan-Anxiety Trial Measured Prescan Anxiety, Not Anxiety During the Scan

The trial’s acute anxiety assessment occurred after CBD dosing but generally before the scheduled cancer scan. It did not directly measure whether CBD reduced distress inside the scanner. The two timing intervals were different Participants received the study drug within 48 hours before a tumor-assessment scan. Anxiety was reassessed two…
