A 2-AG Enzyme Review Distinguished Total Brain Activity From Cellular Location

A review of MAGL, ABHD6 and ABHD12 explained why their relative contribution to total brain 2-AG breakdown does not fully describe their local signaling role. Where an enzyme acts can matter alongside how much activity it contributes.

The review described different positions in the circuit

MAGL was discussed near presynaptic cannabinoid signaling, while neuronal ABHD6 was positioned near postsynaptic 2-AG generation. These locations support different proposed roles in terminating or limiting signals. A whole-brain activity percentage cannot identify every local effect.

Acute and sustained inhibition differed

The synthesis described acute MAGL inhibition increasing 2-AG and cannabinoid-related responses, while sustained inactivation could produce overload, desensitization and tolerance. A short-term increase in signaling does not establish a durable treatment benefit.

Uncertainty remained for another enzyme

The review discussed ABHD12 and its disease association while questioning its precise status within endocannabinoid signaling. A reported hydrolysis contribution does not settle every physiological role. That uncertainty should remain visible rather than making all three enzymes interchangeable targets.

Interpreting the evidence

The source supports interpreting enzyme amount, cellular position and exposure duration together. It is a mechanistic review, not evidence that manipulating any 2-AG enzyme safely reproduces marijuana effects or provides a validated clinical tolerance strategy.

The CBD Scan Trial Used Equal Allocation Despite Planning a Two-to-One Ratio

The scan-anxiety study ultimately randomized twenty-five participants to each treatment. The paper disclosed that its original sample-size plan anticipated a two-to-one allocation instead. The discrepancy concerned the allocation ratio The central registrar inadvertently implemented a one-to-one scheme rather than the originally intended unequal ratio. Participants still underwent randomized assignment, and…

Only Six Participants Had Severe Baseline Anxiety in the CBD Scan Trial

The scan-anxiety trial stratified randomization by baseline anxiety severity, but only six participants were in its severe category. Stratification did not supply enough data for a reliable severe-anxiety subgroup analysis. The strata helped organize allocation Participants were assigned within GAD-7 categories covering mild-to-moderate and severe anxiety. Of the fifty women…

The CBD Scan-Anxiety Trial Measured Prescan Anxiety, Not Anxiety During the Scan

The trial’s acute anxiety assessment occurred after CBD dosing but generally before the scheduled cancer scan. It did not directly measure whether CBD reduced distress inside the scanner. The two timing intervals were different Participants received the study drug within 48 hours before a tumor-assessment scan. Anxiety was reassessed two…
