Delta-10 THC Metabolites: Human Hepatocytes Are Not Confirmed Human Urine Evidence

Human liver-cell experiments suggested that delta-10 THC follows a different metabolic pattern from delta-8 and delta-9 THC. The study did not confirm those delta-10 findings in urine from people known to have consumed it.

The primary model was in vitro

Researchers incubated THC isomers with human hepatocytes and analyzed proposed metabolites using high-resolution mass spectrometry. Human-derived cells provide useful metabolism evidence, but they are not a controlled administration study in people.

The investigators also examined nineteen routine urine specimens containing delta-8 or delta-9 metabolites. No authentic delta-10-positive urine specimen was available. The presence of a human urine component therefore does not make every isomer’s findings clinically confirmed.

Structural assignments had different certainty

Metabolites were proposed from accurate mass, fragmentation, retention behavior and comparisons with available standards or controls. Some glucuronide structures remained tentative, and the routine method was not optimized for every conjugate.

Where reference standards were unavailable, signal abundance could not be assumed to equal molar abundance across different compounds. Different ionization and response factors can affect apparent proportions.

Why this matters for testing

Closely related isomers may share fragments while differing in metabolism. A method validated only for delta-9 metabolites may not identify or quantify another isomer correctly. Hydrolysis also changes the profile being observed by removing conjugates.

That creates a need for validated separation and authentic standards, rather than permission to identify any unfamiliar peak as delta-10 exposure.

What remains to be established

The authors call for confirmation with reference materials and authentic human specimens after delta-10 intake. The experiment did not establish dose, excretion duration, impairment or an individual clinical toxicity profile for delta-10.

The practical conclusion is a qualified one: the laboratory patterns identify promising analytical targets and possible misclassification problems. They do not yet provide a fully validated human exposure marker or a shortcut for interpreting an unconfirmed case result.

For context, see our discussion of THC concentrations and the separate question of impairment.

A Higher Edible Dose Did Not Produce a Higher Mean Blood THC Peak in Every Group

A 50 mg edible THC dose did not produce a higher mean whole-blood THC peak than 25 mg in a small between-group study. That observation does not show reversed absorption or make blood concentration a reliable dose calculator. Only six people received each dose Healthy adults without recent cannabis use…

Oral-Fluid Parent Cannabinoids and Metabolites Followed Different Route Patterns

An oral-fluid THC peak immediately after cannabis use can reflect material deposited in the mouth rather than peak systemic exposure. A controlled comparison of smoked, vaporized and edible cannabis found different route patterns for parent cannabinoids and metabolites. Within-person route comparisons Eleven frequent and nine occasional cannabis smokers completed a…

Transdermal CBD’s Open-Label Seizure Improvement Did Not Reverse Its Blinded Null Result

Transdermal CBD did not significantly reduce seizure frequency compared with placebo during a blinded adult focal-epilepsy trial. Improvement during the later open-label extension answered a different question and did not overturn that randomized comparison. A daily-dose distinction clarified by correction The trial randomized 188 adults to transdermal CBD or placebo….
